Gram staining is the most common method used to distinguish two massive groups of bacteria based on their exclusive cellular wall elements. The Gram stain process distinguishes between Gram-positive and Gram-negative groups by using coloring those cells red or violet. Gram-positive bacteria stain violet due to the presence of a thick layer of peptidoglycan in their cell wall, which retains the crystal violet these cells are stained with. Instead, Gram-negative microorganism stain red, that's attributed to a thinner peptidoglycan wall, which does not keep the crystal violet throughout the decoloring technique.
